一般的な汚染物質で いつの間にか男らしさを失う

7つの新たな動物実験によって、プラスティックに広く用いられている殺虫剤と化学物質が、男性の生殖機能に影響を与えることが発見された。これらの化学物質は、生殖機能の発育をプログラムするかのように、男性の性ホルモンの作用を妨げることで生殖機能に大きなダメージを与える。
プラスティック可塑剤(軟化剤)として、いたるところで使用されている油性溶剤のテレフタレートは、環境中で最も豊富に存在する合成化学物質となっている。塩ビ製品のほとんどに軟化剤として用いられているジエチルヘキシルフタレート(DEHP)や、蚊用防虫剤の添加剤として用いられているジブチルフタレート(DBP)を胎児に曝露した時の影響について、ある1つの新しい研究が行われている。
リサーチトライアングルパーク(Research Triangle Park, N.C.,)にある環境保護庁の L. Earl Gray Jr. らは、雌ラットの母乳中に含まれる化学物質を管理し、母乳が化学物質に曝露していない母親の子供と、200−1,000mg/体重1kgの化学物質を1回投与した雌ラットの子供を調査した。
フタレートに曝露していない母親の子供と比較したところ、曝露した母親の子供はわずかなテストステロンしか作り出さず異常値を示した。また、時折睾丸の1つが欠如し、血嚢のようなものが現れた。Grayは「このような現象を見たのは初めてだ」と言った。
生前にどちらのフタレートに曝露しても、生まれてきた子供の結腸からベニスの付け根に走る筋肉が著しく萎縮した。ある子供では精巣上体(副睾丸)と精子貯蔵組織が正常な大きさではなく、ただの断片にすぎない大きさになっていた。
Grayのチームはこれらの事実の目録を作成した。そして、男性の性ホルモンやアンドロゲンを妨げる化学物質によりこれまで見られた多くの異常についても目録を作成した。実験動物の多くは、女性の特徴を持った乳首をもって生まれた。そのデータは、2週間前にニューオーリンズで開かれた毒物協会の会合で発表された。また、「毒物と産業衛生(1月−3月号)」に掲載された。
リサーチトライアングルパーク(Research Triangle Park, N.C.)にある「毒物化学産業協会」のPaul M.D. Foster らは、フタレートの反アンドロゲン効果について探索研究してきた。彼らは毒物協会の会合において、母親の体重1kg当たりわずか100mgのDBPを投与することで、雄の発育障害が発生すると報告した。
彼らのデータによれば、DBPは胎児の精巣で作られるテストステロンを半減する。Fosterによると、それは動物が大人になった後に、精巣腫瘍を引き起こす増殖細胞を2倍作ることになるのだと。
商業ベースで用いられる多くの化学物質は、反アンドロゲン作用を持っている。「毒物と産業衛生」誌にある他の4つの論文において、Grayのグループは様々な生殖器障害を引き起こす2つの殺菌剤(ビンクロゾリン、vinclozolinやプロシミドン、procymidone)、除草剤(リヌロン、linuron)、殺虫剤(メトキシクロル、methoxychlor),他の化学物質について述べている。
リヌロンが及ぼす破壊的影響は驚くべき状況を示した。通常、外部生殖器官の異常は内部組織が崩壊していることを示す。しかし、リヌロンに曝露した子供は外部奇形がなかったにも関わらず、その50%は、精巣上体(副睾丸)が欠如し精巣奇形になった。とグレーは観察している。彼のグループはまた、思春期直前の若い雄の動物を反アンドロゲン殺虫剤に暴露することによって、生殖器官の成長が劇的に遅延することを示した。
「これらのラットで生殖器障害などの異常が発生したフタレートや殺虫剤の投与量は、人間が実際に曝露しているレベルに近い。」と科学と環境衛生ネットワーク科学担当ディレクターでもあるボストン内科医のTed Schettler は言っている。透析患者やプラスティック製点滴用バッグから点滴液を受けている人々の多くはDEHPに曝露している。歯で噛むような使い方をする塩ビ樹脂製玩具においてもフタレートの曝露を受ける。
リッチモンドにあるバージニアイギリス連邦大学のPeter L. deFur は、「現代人における反アンドロゲン作用を有する化学物質への曝露は、健康に影響を及ぼしていると言ってもいい状況にある。またその影響は把握可能であろう。」と懸念している。
−私のコメント−
フタレート(またはフタル酸エステル)は、接着剤の可塑剤、粘着テープの粘着剤の可塑剤、塩ビ樹脂の可塑剤など非常に多くの用途に用いられている。環境ホルモン作用の疑いがある化学物質の代表格であり、日本国内でも河川や海洋などの環境中から他の環境ホルモン物質やダイオキシン類と比べて多量に検出されている。しかし、塩ビや可塑剤業界は、人間の健康に影響のないレベルだと反論している。
しかし、例え動物実験であっても本論文のような研究結果が報告される限り、私達の健康への影響について真剣に考えなければならないのではないだろうか。私達の次世代の子供達に影響が出てからでは遅いのだ。
